Subculture:
seed out at 1-2 x 106 cells/75 cm2; split confluent culture 1:4 once a week using the double amount of trypsin/EDTA for up to 20 min (cells are difficult to detach); reduced viability immediately after thawing (ca. 50%), cells recover subsequently over the next weeks
Incubation:
at 37 °C with 5% CO2
Doubling time:
ca. 3-4 days
Harvest:
cell harvest of ca. 10-12 x 106 cells/175 cm2
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
