80% Advanced RPMI 1640 + 20% h.i. FBS + 1 mM sodium pyruvate + 2 mM L-glutamine + 4.5 g/L glucose + 50 µM mercaptoethanol
Subculture:
seed out at ca. 0.5 x 106 cells/ml; optimal split ratio of 1:2 to 1:3 every 2-3 days, maintain at 0.5-1.5 x 106 cells/ml

at 37 °C with 5% CO2
