Primary keratinocyte cells isolated from a neonatal foreskin were transduced with replication deficient amphotropic retroviruses containing a) the catalytic subunit of human telomerase reverse transcriptase hTERT, and b) SV40 early region for immortalization; donor, vector, and recipient cells are categorized to biosafety level 1

Subculture:
seed out at ca. 0.3 x 106 cells in a T75 flask; split 1:4 - 1:8 every 5-8 days using Accutase

cell harvest of ca. 2-3 x 106 cells/75 cm2
