Origin:
Genetic modification of HEK293-DR-GFP-RAD51D-16 (DSMZ ACC 847) which belongs to a series of genetically disrupted RAD51 paralogs. Here, in ACC 852, the RAD51D gene is functionally reconstituted by retroviral transfection. Cells are described in the literature to be puromycin and hygromycin resistant

Medium:
90% DMEM + 10% h.i. FBS + (additionally) 2 mM L-glutamine (to sustain genetic stability cells should be maintained, at least periodically, with 0.5 µg/ml puromycin and/or 100 µg/ml hygromycin)
Subculture:
culture vessels should be coated with 0.01% poly-L-lysine solution; seed out at 1 x 106 cells/80 cm2; split 1:10 to 1:20 once or twice a week using TrypleExpress (trypsin/EDTA may not be efficient)
Incubation:
at 37 °C with 5% CO2

ca. 30 hours

cell harvest of ca. 20 x 106 cells/80 cm2 flask
