Origin:
established in 1996 from the tumor tissue of a 57-year-old man with local recurrence of follicular thyroid cancer (containing well-differentiated with dispersed poorly differentiated areas). Cells express the transcriptionally inactive (dominant negativ) delta-p53 isoform which is additionally mutated resulting in a truncated p53 form and a short 5 KDA fragment (delta p53 -/-). A sister cell line with an untruncated delta-p53 is available (ACC 510). Useful for studies with p53 family members p63 and p73

Morphology:
epithelioid cells growing adherently in monolayers
Medium:
80-90% RPMI 1640 + 10-20% h.i. FBS + 1 mM sodium pyruvate + 1x IST (5 µg/ml insulin + 5 µg/ml transferrin + 5 ng/ml sodium selenite)
Subculture:
seed out at about 1 x 106 cells/25cm2; split 1:2 to 1:3 once a week using trypsin/EDTA
Incubation:
at 37 °C with 5% CO2
Doubling time:
ca. 100 hours
Harvest:
cell harvest of ca. 15 x 106 cells/175 cm2 flask
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
