Medium:
90% DMEM + 10% h.i. FBS + 2 mM L-glutamine + 1% sodium pyruvate +1% non-essential amino acids
Subculture:
seed out at about 1 x 106 cells/80 cm2;
split semi-confluent culture up to 1:10 once a week using TripleExpress. Flasks coated with 0.01% poly-L-lysine may be used if the cells do not adhere
Incubation:
at 37 °C with 5-10% CO2
Doubling time:
ca. 45 hours
Harvest:
cell harvest of ca. 10 x 106 cells/80 cm2 flask
