90% MEM (with Earle's salts) + 10% h.i. FBS
Subculture:
seed out at ca. 1 x 106 cells/80 cm2 flask; split confluent culture 1:2 to 1:4 every 4-8 days using trypsin/EDTA for 5-10 min (before trypsination wash 2x with PBS); initially after thawing cells may grow slowly
Incubation:
at 37 °C with 5% CO2
Doubling time:
ca. 60-80 hrs (after recovery from liquid nitrogen ca. 100 hrs)
Harvest:
ca. 15 x 106 cells/175 cm2 flask
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
