Origin:
genetically modified B-lymphoblastoid cell line established in 1993 by infection of cord blood with an experimental EBV system. Cells express an estrogen-receptor-EBNA2 fusion protein (ER/EBNA-2), confering growth dependency by ß-estradiol. ER/EB2-5
is the parental cell line of P493-6 (DSMZ ACC 915)

Risk assessment:
The cell line is positive for EBV (HHV-4) by PCR analysis. However, expression of immediate-early protein BZLF-1 was negative by western blot analysis in untreated and phorbol ester / sodium butyrate stimulated cells. However, the cells are genetically modified with a plasmid containing a ß-estradiol inducible EBNA2 gene to complement the originally deleted gene of EBV.
According to a recommendation of the German Central Committee for Biological Safety (ZKBS) the cells have to be handled under biosafety level 2.

90-95% RPMI 1640 + 5-10% h.i. FBS + 2mM L-glutamine (i.e.total concentration: 4 mM) + 1 µM ß-estradiol
Subculture:
seed out at about 0.2-0.5 x 106 cells/ml; split 1:5 to 1:10 once or twice a week and maintain at 0.4-0.8 x 106 cells/ml
Incubation:
at 37 °C with 5% CO2
Doubling time:
about 24-48 hours, depending on the split ratio
Harvest:
maximal density of ca. 1 x 106 cells/ml

frozen with 70% medium, 20% FBS, 10% DMSO
