Origin:

Morphology:

round (relatively large) cells growing singly in suspension (ca. 1-2% cells are slightly adherent); image; image; image; image; image

Medium:

80-90% alpha-MEM (with ribo- and deoxyribonucleosides) + 10-20% h.i. FBS + 5 ng/ml GM-CSF

Subculture:

seed out at ca. 0.5 x 10⁶ cells/ml; maintain at 0.5-1.0 x 10⁶ cells/ml; cultivate in 12- or 24-well-plate for optimal growth; split saturated culture 1:2 every 2-3 days. CAVEAT: by favoring selective growth, suboptimal culture of cytokine-dependent cell lines may promote outgrowth of factor-independent subclones. Selective conditions include cytokine insufficiency and inadequate cell density. Thus the DSMZ cannot guarantee indefinite stability of factor-dependence of cell lines.

Incubation:

at 37 °C with 5% CO₂

ca. 70 hours

Harvest:

saturation density at ca. 1.0-1.5 x 10⁶ cells/ml

frozen with 70% medium, 20% FBS, 10% DMSO
