AM-C6SC8
ACC 152
Cell line:
AM-C6SC8
DSMZ no.:
ACC 152
Species:
porcine (pig) (Sus scrofa)
Cell type:
normal kidney cells
Origin:
derived as a subclone from the cell line AM II which was established from a normal porcine kidney in 1983; cells have been used in assays to test toxins and mutagens
Reference(s):
Biosafety level:
1
Risk assessment:
Reverse transcripase activity has been detected in this cell line by an RT-ELISA assay. Furthermore, the cells contain DNA sequences and their transcripts of Porcine type-C oncoviruses (PCOV). In Germany, these viruses and cell lines infected with these viruses are classified BSL 1 regarding men and BSL 2 relating to animals. However, if the cells are used in terms of genetic modifications, the cells have to be handled under BSL 2 conditions (see statement of the German Central Committee on Biological Safety - ZKBS - Ref. 6790-10-65)
Permissions and restrictions:

Medium:
80-90% Medium 199 (Hank's salts) + 10-20% h.i. FBS
Subculture:
cells are difficult to culture and grow slowly to very slowly! split confluent culture 1:5 to 1:10 about once a week using trypsin/EDTA
Incubation:
at 37 °C with 5% CO2
Harvest:
cell harvest of about 8 x 106 cells/80 cm2 or 1 x 107 cells/175 cm2
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
DSMZ Scientific Data:
Mycoplasma:
negative in DAPI, microbiological culture, RNA hybridization assays
Species validation:
Species-level identification was carried out by mitochondrial Cytochrome C Oxidase Subunit 1 (COI) DNA barcoding according standard ANSI/ATCC ASN-0003-2015 and revealed Sus scrofa species.
