Origin:
established from the embryonal (stage 52/53) normal heart endothelium (tadpole heart) of a South African clawed frog (Xenopus laevis Daudin) in 1979; cells were described in the literature to synthesize factor-VIII-related antigen (AHG-a.p.), fibronectin and laminin and to be tetraploid (72 +/-5, 2N = 36)

90% DMEM (diluted with 20% distilled water) + 10% h.i. FBS + 20-30 mM HEPES
Subculture:
split confluent culture 1:2 every 10-12 days using trypsin/EDTA; avoid centrifugation, just dilute the harvested cells with medium and seed into a new flask; cells grow extremely slowly with a doubling time of about 10-12 days; thaw in cold water (10 °C), dilute with 10-fold volume of fresh culture medium, centrifuge gently (about 80 x g for 5 min)
Incubation:
keep at room temperature in closed flasks (without exposure to CO2)

ca. 10-12 days
Harvest:
cell harvest of about 2.5 x 106 cells/80 cm2
Storage:
frozen with 90% complete medium + 10% glycerol (not DMSO)

Species validation:
Species-level identification was carried out by mitochondrial Cytochrome C Oxidase Subunit 1 (COI) DNA barcoding according standard ANSI/ATCC ASN-0003-2015 and revealed Xenopus laevis species.
