Origin:
the rat basophlic leukemia (RBL) line was originally obtained by Eccleston et al. and was maintained by serial passage in neonatal Wistar rats; four cell lines (named RBL I-IV) were started from this tumor between 1973 and 1975; the subline RBL-IV HR+ was derived from RBL-IV; further subcloning established the cell line RBL-2H3; cells are described in the literature to release histamine as an IgE-mediated reaction

70% MEM (with Earle's salts) + 20% RPMI 1640 + 10% h.i. FBS; a higher proportion of RPMI 1640 may increase proliferation rate, but decreases histamine production
Subculture:
split culture before confluency 1:5 every 3 days using trypsin/EDTA; seed out at ca. 1-2 x 106 cells/80 cm2
Incubation:
at 37 °C (some researchers favour cultivation with CO2)
Doubling time:
ca. 40--60 hours
Harvest:
cell harvest of ca. 15-20 x 106 cells/175 cm2

frozen with 70% medium, 20% FBS, 10% DMSO

contamination was eliminated with Ciprobay (ciprofloxacin), then negative in DAPI, microbiological culture, RNA hybridization, PCR assays

Species validation:
Species-level identification was carried out by mitochondrial Cytochrome C Oxidase Subunit 1 (COI) DNA barcoding according standard ANSI/ATCC ASN-0003-2015 and revealed Rattus norvegicus species.

rat hypertriploid karyotype - 68(62-72)<3n> - carries large metacentric marker
