BETA-TC-3
ACC 324
Cell line:
BETA-TC-3
DSMZ no.:
ACC 324
Species:
murine (mouse) (Mus musculus)
Cell type:
pancreas insulinoma (beta-tumor)
Origin:
established from pancreatic insulinoma excised from a transgenic mouse carrying hybrid insulin-SV40 large T genes in 1988; described as producing pro-insulin I and II processed into mature insulin (inducible 30-fold by glucose); however, the DSMZ did not verify the production of insulin
Biosafety level:
1
Permissions and restrictions:
DSMZ Cell Culture Data:
Medium:
82.5% DMEM (4.5 g/L glucose) + 15% horse serum + 2.5% h.i. FBS
Subculture:
seed out at ca. 1-4 x 106 cells/175 cm2; split confluent culture 1:5 to 1:10 every 2-5 days by mechanical disruption (trypsin/EDTA can be used for a very short time); the DSMZ cannot guarantee the production of insulin
Incubation:
at 37 °C with 7.5-10% CO2
Doubling time:
ca. 34 hours
Harvest:
cell harvest of ca. 2-3 x 105 cells/cm2
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
DSMZ Scientific Data:
Mycoplasma:
negative in DAPI, microbiological culture, RNA hybridization, PCR assays
Species validation:
Species-level identification was carried out by mitochondrial Cytochrome C Oxidase Subunit 1 (COI) DNA barcoding according standard ANSI/ATCC ASN-0003-2015 and revealed Mus musculus species.
Cytogenetics:
murine hypotetraploid karyotype - 74(70-78)<4n> - 2-4 centric fusion markers and 1-3 dmin present
Viruses:
ELISA: reverse transcriptase positive; PCR: SMRV -
