Medium:
80% Iscove's MDM + 20% h.i. FBS + 10 ng/ml murine IL-3 (or 10-20% conditioned medium of cell line WEHI-3B, DSM ACC 26)
Subculture:
split saturated culture 1:2 every 3-4 days; seed out at ca. 1.0 x 106 cells/ml; after thawing, cells are initially very difficult to culture and viability may be initially rather low (20-30%): hence it is advisable to culture the cells first in 24-well-plates and to exchange the medium, adding fresh mIL-3, already about 5-8 hours after thawing and then daily. CAVEAT: by favoring selective growth, suboptimal culture of cytokine-dependent cell lines may promote outgrowth of factor-independent subclones. Selective conditions include cytokine insufficiency and inadequate cell density. Thus the DSMZ cannot guarantee indefinite stability of factor-dependence of cell lines.
