established in 1993 from the peripheral blood of a 61-year-old man with Philadelphia chromosome negative chronic myeloid leukemia (CML) with trilineage myelodysplasia at refractory leukemic transformation into acute myeloid leukemia (AML M4); carries t(6;9)(p23;q34) leading to DEK-NUP214 (DEK-CAN) fusion gene; contrary to what was originally described the cells do not respond to G-CSF

seed out at ca. 1.0 x 106 cells/ml in 24- or 12-well plates; optimal split ratio of 1:2 or less every 3-4 days; cultures may contain a high amount of cell debris; the accumulation of cell debris can be prevented by using new culture vessels every second week
