Morphology:
neuronal-like cells growing adherently as single cells with tendency to form clusters; FBS-induced differentiation may be visible by predominant monolayer formation
Medium:
Neurobasal medium + 0.5x B-27 supplement with vitamin A, 0.5x N-2 supplement, 20 ng/ml bFGF, 20 ng/ml EGF, 2 mM L-glutamine; omit FBS!
Subculture:
seed out at ca. 1.0 x 106 cells/25 cm2 flask; split culture 1:2 to 1:4 once or twice a week; trypsin should not be used, but cells can be detached by tapping the flask or vigorous pipetting
Incubation:
at 37 °C with 5% CO2
Doubling time:
ca. 3-5 days
Harvest:
cell harvest of ca. 8-10 x 106 cells/80 cm2
Storage:
frozen with 90% medium, 10% DMSO (no FBS added)
