80% mixture of DMEM and Keratinocyte SFM (at 2:1) + 20% h.i. FBS
Subculture:
seed out at ca. 1-2 x 106 cells/80 cm2 flask; split semi-confluent culture 1:2 to 1:5 once or twice a week using trypsin/EDTA or TrypLE Ex for about 10 min (cells are difficult to detach; rinse 2x with PBS before adding trypsin)
Incubation:
at 37 °C with 5% CO2

ca. 2-3 days
Harvest:
cell harvest of 5-8 x 106 cells/175 cm2
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
