Origin:
established from malignant ascites of a metastatic lobular breast cancer of a 72-year-old woman after chemotherapy (tamoxifen) and radiotherapy; cells are described in literature to carry a CDH1 mutation (lack of E-Cadherin expression). Primary tumor cells were ER-positive but others suggested as triple-negative. Due to a mutation in the TP53 gene cells are temperature sensitive. Further described to overexpress the MDR1/ABCB1 drug transporter, to lack beta-catenin expression, to be hyperdiploid and tumorigenic in SCID mice; RNA sequence data are available (see Ref 40906 and RNA-Seq)

80% RPMI 1640 + 20% h.i. FBS, containing additionally 10 mM HEPES, 2.5 g/l glucose, 2 mM L-glutamine, 1 mM pyruvate, 10 µg/ml insulin
Subculture:
seed out at ca. 1 x 106 cells/80 cm2; split confluent culture 1:4 to 1:8 every 5-10 days using trypsin/EDTA; due to the temperature sensitivity cells should be kept
and handled whenever possible at 37°C; medium should be pre-warmed before use
Incubation:
obligatory at 37 °C with 5% CO2
Doubling time:
in the first 1-2 weeks about 200 hours then 80-120 hours
Harvest:
ca. 15-20 x 106 cells/80 cm2
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
