Origin:
Derivative of the modified human embryonal kidney cell line 293 Tet-Off (Clontech) as reporter cell line for studying chromosomal double-strand repair (DR). The cell line contains the hprtDR-GFP reporter and is puromycin resistant. A series of modified cell lines with disruptions in RAD51 paralogs are also available (DSMZ ACC 845-ACC 854).

Medium:
90% DMEM + 10% h.i. FBS + (additionally) 2 mM L-glutamine (to sustain genetic stability cells should be maintained, at least periodically, with 0.5 µg/ml puromycin)
Subculture:
culture vessels should be coated with 0.01%
poly-L-lysine solution; seed out at 1 x 106 cells/25 cm2; split subconfluent culture 1:3 to 1:4 every 2-3 days using trypsin/EDTA; optimal density around 4 x 106 cells/25 cm2
Incubation:
at 37 °C with 10% CO2
Doubling time:
ca. 24 hours
Harvest:
cell harvest of ca. 8 x 106 cells/25 cm2 flask
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
