Origin:
established in 2014 from a cutaneous metastasis of a 48-year-old man with highly aggressive ulcerated melanoma after chemotherapy. Cells are described to be moderately hyperdiploid, carry a mutation in the N-Ras-gene (pQ61R) but not in the BRAF. Further described to be tumorigenic in nude mice and able to grow in 3D using hydrogel

elongate or triangular dendritic cells growing adherently as monolayer; cells are highly pigmented and sedimented cells have a brown color
Medium:
80% RPMI 1640 + 20% h.i. FBS + 2 mM glutamine
Subculture:
seed out at 1 x 106 cells/80 cm2;
split confluent culture 1:4 to 1:6 every 4-6 days using trypsin/EDTA
Incubation:
at 37 °C with 5% CO2

ca. 48 hours

cell harvest of ca. 5 x 106 cells/80 cm2 flask
