Medium:
80% MEM (with Earle's salts) + 20% h.i. FBS + non-essential amino acids + 2 mM L-glutamine
Subculture:
seed out at about 1 x 106 cells/80 cm2; split semiconfluent culture 1:5 once a week using trypsin/EDTA (wash 2x with PBS before adding the trypsin)
Incubation:
at 37 °C with 5% CO2
Doubling time:
ca. 60-70 hours
Harvest:
cell harvest of ca. 5 x 106 cells/80 cm2 flask
