Medium:
80% MEM (with Earle's salts) + 20% h.i. FBS + 2 mM L-glutamine
Subculture:
seed out at about 1-2 x 106 cells/80 cm2; split confluent culture 1:3 to 1:5 every 2-3 days using trypsin/EDTA (wash 2x with PBS and then incubate for up to 10 min with trypsin)
Incubation:
at 37 °C with 5% CO2
Doubling time:
ca. 48 hours
Harvest:
cell harvest of ca. 8-10 x 106 cells/80 cm2 flask
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO
