Medium:
95% Leibovitz´ L15 (without phenol red) + 5% (not inactivated) FBS
Subculture:
cells grow very slowly; seed out 1-1.5 x 106 cells in a 25 cm2 flask; split confluent culture 1:2 every 10-14 days as follows: rinse 2x with 1ml versene (0.2 g/l) add 0.6-0.8 ml trypsin (0.25% (for 1-2 min)) and detach carefully; avoid sheering the cells or formation of foam; change medium every 5-7 days
Incubation:
at ca. 20°C in the dark, without CO2 gas exchange
Doubling time:
ca. 10-12 days
Harvest:
cell harvest of 8-10 x 106 cells/80 cm2 flask
Storage:
frozen with 70% medium, 20% FBS, 10% DMSO

Mycoplasma:
contamination was eliminated with MRA and BM-cyclin by the originator, then proofed as negative by PCR at the DSMZ
Species validation:
Species-level identification was carried out by mitochondrial Cytochrome C Oxidase Subunit 1 (COI) DNA barcoding according standard ANSI/ATCC ASN-0003-2015 and revealed Oncorhynchus mykiss species.
